outcomes for children with leptin signalling deficiency who do not receive treatment for the condition. This retrospective cross-sectional study included 145 children from the Punjab region


of Pakistan, with recruitment beginning in 2012. The children all had mutations that caused deficiency of leptin, leptin receptor or melanocortin 4 receptor. Physical, clinical and metabolic


evaluations were conducted. The children with leptin or leptin receptor deficiency tended to have more severe hyperphagia and earlier onset of excessive weight gain than the children with
